189 Integrating single-cell and spatial transcriptomics of human hair follicles to define transcriptional signature of follicular dermal papilla

چکیده
The follicular dermal papilla (DP) is a specialized hair mesenchymal cell that plays pivotal roles in follicle (HF) development and morphogenesis. Although transcriptome data exists for HF, characterization of human DPs at the single-cell level has been unsatisfactory due to limited number DP cells under dissociation condition. To generate high proportion cells, we manually isolated anagen HFs from three scalp specimens used these populations RNA sequencing (scRNA-seq). In addition, Digital Spatial Profiling (DSP) provide expression profile with spatial information. Using computational deconvolution methods, integration scRNA-seq DSP estimated epithelial abundance hair-specific regions interest confirm utility identifying molecular markers mesenchyme. Analysis HF revealed RSPO4, WNT5A, TRPS1, CRABP1, APCDD1, EDN3, RSPO3, LEF1, BMP4, BMP7, NRG2 are specifically expressed whereas MME, LGR4, LGR5, FZD1, CXCL14, TAGLN were mainly sheath. markers, performed situ hybridization (RNA-ISH). RNA-ISH, observed presence MSX1 HOXC13 mRNA matrix BMP5, DKK3, RSPO4 LGR6 DP. Collectively, through transcriptomic profiling, could establish foundation understand characteristics

Herbal Extracts Induce Dermal Papilla Cell Proliferation of Human Hair Follicles
BACKGROUND The number of people suffering from balding or hair thinning is increasing, despite the advances in various medical therapies. Therefore, it is highly important to develop new therapies to inhibit balding and increase hair proliferation. OBJECTIVE We investigated the effects of herbal extracts commonly used for improving balding in traditional medicine to identify potential agents ...
متن کاملhair follicle generation by injecting adult human follicular epithelial and dermal papilla cells in nude mice
objective: objective: dermal papilla and hair epithelial stem cells regulate hair formation and growth cycle. damage to or losing of these cells can cause hair loss. although several studies have claimed to reconstitute hairs using rodent cells in animal model, a stable human hair follicle reconstitution protocol needs more studies. here in this study we evaluated hair induction through injecti...

The dermal papilla is a cluster of mesenchymal cells located at the base of the hair follicle which have a number of important roles in the regulation of hair growth. As a consequence, in vitro models of these cells are widely used to study the molecular mechanisms which underlie hair follicle induction, growth and maintenance. While dermal papilla from rodent hair follicles can be digested pri...
متن کاملDermal papilla cell number specifies hair size, shape and cycling and its reduction causes follicular decline.
Although the hair shaft is derived from the progeny of keratinocyte stem cells in the follicular epithelium, the growth and differentiation of follicular keratinocytes is guided by a specialized mesenchymal population, the dermal papilla (DP), that is embedded in the hair bulb. Here we show that the number of DP cells in the follicle correlates with the size and shape of the hair produced in th...
متن کاملMorphogenesis of chimeric hair follicles in engineered skin substitutes with human keratinocytes and murine dermal papilla cells.
Engineered skin substitutes (ESS) have been used successfully to treat life-threatening burns, but lack cutaneous appendages. To address this deficiency, dermal constructs were prepared using collagen-glycosaminoglycan scaffolds populated with murine dermal papilla cells expressing green fluorescent protein (mDPC-GFP), human dermal papilla cells (hDPC) and/or human fibroblasts (hF). Subsequentl...
